Enumeration Techniques of Newcastle Disease Virus (NDV) for Oncolytic Virotherapy

چکیده
The present work was carried out to evaluate different quantitation techniques when Newcastle disease virus was intended to use as an oncolytic agent. The R 2 B Mukteshwar strain of NDV was procured as lyophilized vaccine. Then application were carried out like haemagglutination test, tissue culture infective dose-50 (TCID50), plaque forming unit (PFU) calculation and real time PCR to enumerate the number of viruses. The HA titre was obtained as 1:128 across the dilutions. While TCID 50 and PFU counts were obtained as 1×108.16/ ml and 4.2×107 PFU/ ml, respectively. Based upon comparison with standard NDV RNA, Real time PCR also revealed the number of virus 108/ ml. HA was found consistent but indirect; contrastingly TCID50 suffered with subjectivity of interpretation. PFU counts were found within a range and lease possibility of interpretation error than TCID 50 . Though real time was found automated, highly specifi c and sensitive assay but handling of RNA and cost were limiting factors.

Immunobiology of Anticancer Virotherapy With Newcastle Disease Virus in Cancer Patients
Virotherapy with oncolytic viruses that preferentially infect and kill cancer cells is a novel and promising strategy for cancer treatment. Newcastle disease virus (NDV), which is pathogenic in birds, has beneficial clinical effects in cancer patients. NDV virotherapy is safe and elicits an antitumor response in patients affected by different types of cancers. The selective replication of NDV i...
متن کاملTherapeutic potential of oncolytic Newcastle disease virus: a critical review
Newcastle disease virus (NDV) features a natural preference for replication in many tumor cells compared with normal cells. The observed antitumor effect of NDV appears to be a result of both selective killing of tumor cells and induction of immune responses. Genetic manipulations to change viral tropism and arming the virus with genes encoding for cytokines improved the oncolytic capacity of N...
متن کاملCross-infection of tumor cells by contact with T lymphocytes loaded with Newcastle disease virus.
Oncolytic virotherapy, a new type of cancer therapy involving viruses with oncolytic and immunostimulatory potential, is based on tumor selective viral replication, resulting in a specific lysis of tumor cells. Effective tumor targeting of oncolytic viruses remains a major problem because only a fraction of systemically applied viruses can reach the tumor tissue. We describe for the first time ...
متن کاملOncolytic Newcastle Disease Virus as Cutting Edge between Tumor and Host
Oncolytic viruses (OVs) replicate selectively in tumor cells and exert anti-tumor cytotoxic activity. Among them, Newcastle Disease Virus (NDV), a bird RNA virus of the paramyxovirus family, appears outstanding. Its anti-tumor effect is based on: (i) oncolytic activity and (ii) immunostimulation. Together these activities facilitate the induction of post-oncolytic adaptive immunity. We will pre...

Hematological malignancies are important diseases that need more powerful therapeutics. Even with current targeting therapies, such as rituximab and other chemotherapeutic agents, there is a need to develop new treatment strategies. Combination therapy seems the best option to target the tumor cells by different mechanisms. Virotherapy is a very promising treatment modality, as it is selective,...
